Applications of BBa_K1075025

We used this part as a IPTG regulatable reporter for investigating SspB inducable degradation systems in E. coli. For initial assays we tested the effect of different IPTG concentrations on mCherry-ssrA expression levels.

IPTG induced mCherry-ssra expression We inoculated 3x 6ml LB with a overnight grown bacterial culture. We let it grow till an OD of about ~0.8 and used the first uninduced culture as a negative control while we induced the other two cultures with different IPTG concentrations (0.2µM and 1µM). After Overnight growth we pelleted 2ml of each culture through centrifugation. The strength of red color indicates mCherry-ssrA epression.
